原理介绍

无铜点击化学反应,即菌株促进的叠氮炔环加成(SPAAC)是一种生物正交反应,利用一对试剂,即环辛基和叠氮化合物,它们彼此有效地反应,同时对羟基、氨基和硫醇等天然官能团保持惰性。SPAAC能够通过形成稳定的三唑,在水和其他复杂的化学环境中标记各种各样的生物分子,而无需任何辅助试剂。在众多已知的环辛基化合物中,二苯并环辛基化合物(DBCO)是一类具有相当快的动力学和良好的水缓冲稳定性的试剂。在生理温度和pH范围内,DBCO基团不会与许多生物分子中天然存在的巯基、氨基或羟基发生反应。DBCO-PEG4-琥珀酰亚胺酯是一种常用的通过氨基修饰将DBCO基团引入生物分子的试剂。靶点科技分销的ClickChemistryTools的DBCO-PEG4-NHS Ester,是高效率的DBCO 琥珀酰亚胺酯。
